Production of Anti-species Complement C3(¥â1c,¥â1A) Globulin in Ascitic Fluid of Mice against Zymosan-C3-Complex

Abstract
Mouse anti¡©species C3(¥â1c,¥â1A) ascitic globulin has been widely used for the demonstration of antigen, antibody or immune complexes by fluorescence anti¡©complement(C3) technique. However, an attempt to use mouse anti¡©species C3 ascitic globulin as antibodies for quantitative study of C3 globulin level in the sera by single radial immunodiffusion test(SRID) has not been made mainly because the yields of immunoglobulin in the mice were far more less than that in other animals. In the present study, a production of ascitic immunoglobulin against guinea pig or human C3 globulin(zymosan¡©C3 complex) in five inbred strains of mice was investigated. In addition, the sensitivity and specificity of the ascitic immunoglobulin as an antibody for the quantitative study of serum complement C3 globulin(¥â1c,¥â1A) by SRID was described. The mouse anti¡©species C3 ascitic fluid began to develop 28 days and the maximum yield appeared 35 days of immunization. Although the total yields of ascitic fluid obtained from 10 mice of ICR¡©IVR, Anyang, C_3H, KK and Black/65 were not significant, the highest titres of precipitating antibody 1:16 was observed in ascitic fluid of ICR¡©IVR mice and C_3H mice. The total yields of ascitic fluid varied from 3 to 10 ml per mouse. The maximum yields of ascitic fluid obtained from 7 of 10 immunized ICR¡©IVR mice was 36.5ml. In electromobility, the mouse anti¡©species C3 ascitic globulin gave a monospecific preciptation band corresponding to ¥â1c globulin. In the quantitative study, by means of SRID, of average levels of serum C3 globulin of 29 in¡©patients¡¯ sera obtained from the National Veterans Administration Hospital, there observed no significant difference between agar gel made of mouse anti¡©human C3 globulin(138.5¡¾47.7§·/100ml) and agar gel of Hyland products(140.3¡¾38.5§·/100ml). These results indicated that the mouse anti¡©species C3¡©ascitic globulins produced by simple intraperitoneal inoculation with zymosan¡©C3 globulin complex are of value as antibodies for SRID in sensitivity and specificity as well as for fluorescence anti¡©complement technique.
